Gonadorelin

Injection, Powder, for Solution · Parenteral · 100 µg

List code
604
Pharmacologic class
Hypothalamic hormone; gonadotrophin-releasing hormone (GnRH, LHRH)

What it is and how it works

Gonadorelin is synthetic GnRH, identical to the hypothalamic decapeptide. Given as a single dose it stimulates release of LH and, to a lesser extent, FSH from the anterior pituitary, allowing assessment of pituitary gonadotrophin reserve; given in physiological pulses it restores normal gonadotrophin secretion, whereas continuous exposure desensitises the pituitary.

Indications

Diagnostic assessment of anterior pituitary gonadotrophin function (GnRH stimulation test) in suspected hypogonadism or delayed puberty; pulsatile administration for induction of ovulation in hypothalamic amenorrhoea, where available.

Dosage and administration (summary)

Diagnostic test: usually 100 micrograms by intravenous (or subcutaneous) injection, with blood samples for LH and FSH taken before and at intervals after the dose. Pulsatile therapy is given by programmable pump under specialist supervision.

Contraindications

Hypersensitivity to gonadorelin; for pulsatile therapy, conditions aggravated by pregnancy, ovarian cysts and hormone-dependent tumours.

Warnings and precautions

Hypersensitivity reactions, including anaphylaxis, have been reported, particularly after repeated administration. During ovulation induction, multiple pregnancy and ovarian hyperstimulation may occur, and local reactions or infection at the pump site should be monitored.

Pregnancy

Not indicated for diagnostic use in pregnancy.

Storage and supply

Store below 25 °C, protected from light (or refrigerated according to the product information).
