What it is and how it works
Idoxuridine is an iodinated thymidine analogue that is phosphorylated and incorporated into viral DNA in place of thymidine. This produces faulty DNA and inhibits replication of herpes simplex virus.
Indications
Topical treatment of herpes simplex keratitis (dendritic corneal ulcer); now largely superseded by aciclovir, ganciclovir or trifluridine.
Dosage and administration (summary)
Instil 1 drop of 0.1% solution into the affected eye every hour by day and every 2 hours at night, reducing frequency after improvement and continuing for a few days after healing.
Contraindications
Hypersensitivity to idoxuridine or iodine-containing preparations; concomitant use with corticosteroids is generally avoided in epithelial herpetic keratitis.
Warnings and precautions
May cause irritation, punctate keratopathy, follicular conjunctivitis and delayed epithelial healing with prolonged use; treatment should not exceed about 3 weeks. Not effective against stromal disease or non-herpetic infections.
Pregnancy
Avoid in pregnancy; teratogenic in animal studies.
Storage and supply
Store at 2–8 °C, protected from light.
