What it is and how it works
Meclozine is a piperazine H1-receptor antagonist with central antimuscarinic and mild sedative properties. It depresses labyrinthine excitability and vestibular–cerebellar conduction, reducing nausea, vomiting and vertigo.
Indications
Prevention and treatment of nausea, vomiting and dizziness of motion sickness; vertigo associated with vestibular disorders.
Dosage and administration (summary)
Motion sickness: 25–50 mg orally about 1 hour before travel, repeated every 24 hours if needed. Vertigo: 25–100 mg daily in divided doses.
Contraindications
Hypersensitivity to meclozine.
Warnings and precautions
Drowsiness may impair driving and is increased by alcohol and other CNS depressants. Antimuscarinic effects warrant caution in angle-closure glaucoma, urinary retention, prostatic hypertrophy, pyloroduodenal obstruction and asthma. Use with caution in the elderly and in hepatic or renal impairment.
Pregnancy
Has been widely used for nausea in pregnancy without evidence of teratogenicity, but use only if needed.
