Interferon Gamma-1b

What it is and how it works

Interferon gamma-1b is a recombinant type II interferon that activates macrophages and phagocytes, enhancing oxidative (respiratory burst) killing of microorganisms, antigen presentation and cytokine production. In osteopetrosis it is thought to enhance osteoclast superoxide production and bone resorption.

Indications

Reduction of the frequency and severity of serious infections in chronic granulomatous disease; delaying progression of severe malignant osteopetrosis.

Dosage and administration (summary)

50 micrograms/m² by subcutaneous injection three times weekly in patients with body surface area above 0.5 m² (1.5 micrograms/kg in smaller patients); dosing is supervised by a specialist.

Contraindications

Hypersensitivity to interferon gamma, closely related interferons or excipients (including E. coli-derived proteins).

Warnings and precautions

Flu-like symptoms (fever, headache, chills, myalgia) are common and may exacerbate pre-existing cardiac disease; caution in patients with seizure disorders, CNS disease or myelosuppression. Monitor blood counts, liver function and renal function periodically. Avoid concomitant myelosuppressive drugs where possible.

Pregnancy

Use only if the potential benefit justifies the potential risk.

Storage and supply

Store at 2–8 °C; do not freeze; do not shake.
